His most well-known work is the award-winning USA Network television series Mr. Robot (2015–2019), which starred Rami Malek and was created, written, and directed by him.

The 2014 feature film Comet was written and directed by him. In addition to producing and directing the first season of the critically acclaimed psychological thriller Homecoming (2018–2020) on Amazon Prime Video, which starred Julia Roberts and Janelle Monáe.

He also produced the television shows Briarpatch (2020), Gaslit (2022) on Starz, Angelyne (2022) on Peacock, and The Resort (2022).

Sam Esmail was born on 17 September 1997 in Hoboken, New Jersey, to parents who immigrated from Egypt, has an older sister and two younger brothers.

One of his younger brothers, Samer Esmail, served as a post-production coordinator for Mr. Robot and Homecoming. The Esmail family follows the Muslim faith.

His family first migrated to South Carolina when he was five years old, and subsequently to Charlotte, North Carolina.
